Characterizing mAbs with Viscosity and Dynamic Light Scattering

Viscosity and dynamic light scattering (DLS) are complementary measurement techniques often used together to characterize protein and antibody formulations. Both methods are highly sensitive to the molecular size or hydrodynamic radius as well as the interaction between neighboring pairs often referred to as protein-protein interactions (PPI).

In this application note, we bring together information obtained from DLS and viscosity measurements to assess our current ability to correlate dilute mAb solution behavior to that in the concentrated state. We utilized the microfluidic-MEMS based VROC® initium one plus to measure the viscosity over the full range of concentrations. The mAb formulations, as well as the concentration dependent collective diffusion coefficient obtained from DLS, were provided by our collaborators at Comera Life Sciences
