Can Proteins Degrade Polymers?

Hyaluronidase is a naturally occurring enzyme that can degrade hyaluronic acid (HA) into oligomers and other smaller molecular weight units. A popular use of hyaluronidase is to break down HA in fillers used in aesthetic practices, which facilitates HA removal and absorption into the body. FDA-approved applications include using hyaluronidase for accelerating the absorption of drugs. Because viscosity correlates with molecular weight, it can be used to characterize the extent of degradation of HA into oligomers when in the presence of hyaluronidase.

For this application note, we used the m-VROC® II viscometer to repeatedly measure the viscosity of one volume of HA sample over multiple hours with or without added hyaluronidase. This allowed us to characterize the extent of enzymatic degradation of HA in the presence of hyaluronidase.
