Application Brief: Extensive Viscosity Characterization of High Concentration Immunoglobulin with 15 microliters!

The next generation m-VROC® II with advanced features and superior repeatability is the ideal viscometer for measuring biological samples such as proteins and mAbs.

A single 15 microliter sample of 262 mg/mL Bovine Gamma Globulin (BγG) in Na phosphate/NaCl buffer was measured over a range of shear rates and at temperatures of 4, 25, and 37 ºC. The testing procedure was as follows: 

  1. 15 microliters of (BγG) was back loaded into the 100 μL test syringe
  2. The m-VROC II Unit was equipped with a B05 chip to perform multiple shear rate and temperature measurements on the sample with five repeats at each shear rate, retrieving the sample when required. 
  3. Chip Cleaning Station (CCS) performed self-cleaning at the end of the test with preset protocols.

The (BγG) sample exhibits Newtonian behavior across the range tested, with an average viscosity of 296 cP at 4 ºC, 65 cP at 25 ºC, and 31 cP at 37 ºC. All error bars correspond in length to three times the standard deviation and are smaller than the size of the symbols.
